These mRNAs are GM-CSF, Pth, and TGFβ and each of them have AREs, or AU-rich cis-elements. The ARE binding protein KSRP has a Pin1 binding site. Pin1 binds to this site and dephosphorylates the serine and isomerizes the peptide bond between Ser181 and Pro182. This isomerization causes the decay of Pth mRNA. KSRP, and other ARE binding proteins like AUF1, are thought to affect the other mRNAs through mechanisms similar to Pth, with the requirement of a phosphorylated serine bonded to a proline in a specific conformation. Pin1 also triggers proline isomerization of Stem-Loop Binding Protein (SLBP), allowing it to control the dissociation of SLBP from histone mRNA. This leads to Pin1 being able to affect histone mRNA decay. == Licensing and regulation == In the European Union, in the scope of medical devices, the European Court places them in some relation to a specific medical purpose: treating a disease or illness. The Court held that regulating devices that do not explicitly manifest medical benefits as medical devices would run counter to the rationale and lead to overregulation. "The legal foundation for regulating medical devices in the EU is the MDR (Regulation (EU) 2017/745), which was adopted in 2016 and came into effect in 2021. The MDR stipulates that for non-medical devices, the 'requirement to demonstrate a clinical benefit (…) shall be understood as a requirement to demonstrate the performance of the device' (Article 61 para 9 MDR). In other words, devices must demonstrate that they function in the way claimed by manufacturers, instead of showing a therapeutic benefit. Moreover, with respect to the safety of non-medical Annex XVI devices, the MDR lays down the following: For the devices referred to in Annex XVI, the general safety requirements (…) shall be understood to mean that the device, when used under the conditions and for the purposes intended, does not present a risk at all or presents a risk that is no more than the maximum acceptable risk related to the product's use which is consistent with a high level of protection for the safety and health of persons(MDR, Annex I, Article 9). In 1948, the Australian psychiatrist John Cade discovered the usefulness of lithium salts in treating mania while working at the Bundoora Repatriation Mental Hospital in Melbourne. He was injecting guinea pigs with urine extracts taken from manic patients in an attempt to isolate a metabolic compound which might be causing mental symptoms. Since uric acid in gout was known to be psychoactive, he needed soluble urate for a control. He used lithium urate, already known to be the most soluble urate compound, and observed that it caused the guinea pigs to become tranquil. Cade traced the effect to the lithium ion itself, and after Cade ingested lithium himself to ensure its safety in humans, he proposed lithium salts as tranquilizers. In 1949, he published his findings in the Medical Journal of Australia in a paper entitled "Lithium salts in the treatment of psychotic excitement". He soon succeeded in controlling mania in chronically hospitalized patients with lithium. This was one of the first successful applications of a drug to treat mental illness, and it opened the door for the development of medicines for other mental problems in the next decades. In 1950, one of Cade's first patients died of lithium toxicity after Cade had successfully treated him and released him from the hospital.
