Many diagnostic approaches depend on microbiological culture to isolate a pathogen from the appropriate clinical specimen. In a microbial culture, a growth medium is provided for a specific agent. A sample taken from potentially diseased tissue or fluid is then tested for the presence of an infectious agent able to grow within that medium. Many pathogenic bacteria are easily grown on nutrient agar, a form of solid medium that supplies carbohydrates and proteins necessary for growth, along with copious amounts of water. A single bacterium will grow into a visible mound on the surface of the plate called a colony, which may be separated from other colonies or melded together into a "lawn". The size, color, shape and form of a colony is characteristic of the bacterial species, its specific genetic makeup (its strain), and the environment that supports its growth. Plates may contain substances that permit the growth of some bacteria and not others, or that change color in response to certain bacteria and not others. Hence, the bacteria initially migrate and assemble at the air interface, followed by the excretion of bacterial cellulose after about 2 days. The mixed, presumably mutualistic culture has been further described as being lichenous, in accord with the reported presence of the known lichenous natural product usnic acid, though as of 2015, no report appears indicating the standard cyanobacterial species of lichens in association with kombucha fungal components.

=== Phylogeny === It is difficult to reconstruct the early stages in the evolutionary "family tree" of animals using only morphology (their shapes and structures) because of the large differences between the major groups of animals. Hence, reconstructions now rely almost entirely on molecular phylogenetics, which groups organisms based on their biochemistry, most commonly by analyzing DNA or RNA sequences. In 1866, it was proposed that Cnidaria and Ctenophora were more closely related to each other than to Bilateria and formed a group called Coelenterata ("hollow guts") because both rely on the flow of water in and out of a single cavity for feeding, excretion and respiration. In 1881, it was proposed that Ctenophora and Bilateria were more closely related to each other, since they shared features that Cnidaria lack, such as a middle layer of cells (mesoglea in Ctenophora, mesoderm in Bilateria) between the outer and inner layer found in other animals. However, more recent analyses indicate that these similarities were evolved independently in both lineages, instead of being present in their common ancestor. The current view is that Cnidaria and Bilateria are more closely related to each other than either is to Ctenophora. This grouping of Cnidaria and Bilateria has been labelled "Planulozoa", named so because the earliest Bilateria were probably similar to the planula larvae of Cnidaria.

Like gars, bowfin are bimodal breathers—they have the capacity to breathe both water and air. Their gills exchange gases in the water, allowing them to breathe, but they also have a gas bladder that serves to maintain buoyancy and also allows them to breathe air by means of a small pneumatic duct connected from the foregut to the gas bladder. === Nomenclature === Standardized GST nomenclature first proposed in 1992 identifies the species to which the isozyme of interest belongs with a lower-case initial (e.g., "h" for human), which precedes the abbreviation GST.
