== History == Cortisone was first identified by the American chemists Edward Calvin Kendall and Harold L. Mason while researching at the Mayo Clinic. Both Reichstein and the team of O. Wintersteiner and J. Mason's contributions to the crystallization and characterization of the compound have generally been forgotten outside of the Mayo Clinic. == Structure and properties == The molecular weight of lactate racemase differs in the various organisms in which it has been found, ranging from 25,000 to 82,400 g/mol. The structure of the enzyme from L. plantarum was solved by Jian Hu and Robert P. Hausinger of Michigan State University and co-workers there and elsewhere. The protein contains a previously unknown covalently-linked nickel-pincer nucleotide (NPN) cofactor (pyridinium 3-thioamide-5-thiocarboxylic acid mononucleotide), where the nickel atom is bound to C4 of the pyridinium ring and two sulfur atoms. This cofactor participates in a proton-coupled hydride-transfer mechanism. There have been a number of recent studies on NPN cofactor synthesis by the LarB, LarE, and LarC proteins. LarB is a carboxylase/hydrolase of nicotinamide adenine dinucleotide (NAD), providing pyridinium-3,5-dicarboxylic acid mononucleotide and adenosine monophosphate (AMP). LarE is an ATP-dependent sulfur transferase that converts the two substrate carboxyl groups into thioacids by sacrificing the sulfur atoms of a cysteine residue in the protein. Finally, LarC inserts nickel into the organic ligand by a CTP-dependent process to complete synthesis of the NPN cofactor.
=== Substrate-like inhibitors === Substrate-like inhibitors (Figure 4) are more common than the non-substrate-likes. They bind either covalently or non-covalently and have a basic structure where the P1-substituent occupies the S1-pocket and the P2-substituent occupies the S2-pocket. Usually they contain a proline mimetic that occupies the S1-pocket. Large substituents on the 2-cyanopyrrolidine ring are normally not tolerated since the S1-pocket is quite small. Since DPP-4 is identical with the T-cell activation marker CD26 and DPP-4 inhibitors are known to inhibit T-cell proliferation, these compounds were initially thought to be potential immunomodulators. When the function against type 2 diabetes was discovered, the cyanopyrrolidines became a highly popular research material. A little later vildagliptin and saxagliptin, which are the most developed cyanopyrrolidine DPP-4 inhibitors to date, were discovered.
