== Legal status == It is scheduled as Class I drug in Canada. Class I classifications is for drugs that have no approved use in humans and poses a high risk for abuse. Carfentanil has been controlled in China since 1 March 2017. The China–United States trade war has included controversy over the effectiveness of this control. In Germany, carfentanil and its stereoisomers and salts are controlled by the Betäubungsmittelgesetz as a Anlage I substance and can only be used with the special permission of the authorities. Carfentanil is classified as Schedule II under the Controlled Substances Act in the United States with a DEA ACSCN of 9743 and a 2016 annual aggregate manufacturing quota of 19 grams (less than 0.7 oz.). Carfentanil requires approval from the Drug Enforcement Agency for veterinary use. Carfentanil has been specifically controlled as a Class A drug in the United Kingdom since 1986.
== Adverse effects == Common side effects include pneumonia and urinary tract infections. Severe side effects may include blood clots or cardiac arrest. Andexanet alfa has a boxed warning that it is associated with arterial and venous blood clots, ischemic events, cardiac arrest, and sudden deaths.

The aim of the week is to increase global awareness of antibiotic resistance. It also wants to promote the correct usage of antibiotics across all fields to prevent further instances of antibiotic resistance. World Antibiotic Awareness Week has been held every November since 2015. For 2017, the Food and Agriculture Organization of the United Nations (FAO), the World Health Organization (WHO) and the World Organisation for Animal Health (OIE) are together calling for responsible use of antibiotics in humans and animals to reduce the emergence of antibiotic resistance. United Nations In 2016, the Secretary-General of the United Nations convened the Interagency Coordination Group (IACG) on Antimicrobial Resistance. The IACG worked with international organizations and experts in human, animal, and plant health to create a plan to fight antimicrobial resistance. Their report released in April 2019 focuses on the seriousness of antimicrobial resistance and the threat it poses to world health. == Chemistry == Tiagabine, also known as (–)-(R)-1-[4,4-bis(3-methyl-2-thienyl)-3-butenyl]nipecotic acid, is a GABA analogue and a derivative of nipecotic acid. Being a nipecotic acid derivative, introduction of 4,4-diphenylbut-3-enyl and 4,4-bis(3-methylthiophene-1-yl)but-3-enyl side chain increased lipophilicity compared to the parent compound, allowing blood–brain barrier permeability and GABA transporter 1 (GAT-1) selectivity. The experimental log P of tiagabine is 2.6. Analogues of tiagabine include CI-966, NNC-711, and SKF-89976A, among others.

=== Surgery === If the TIA affects an area that is supplied by the carotid arteries, a carotid ultrasound scan may demonstrate stenosis, or narrowing, of the carotid artery. For people with extra-cranial carotid stenosis, if 70-99% of the carotid artery is clogged, carotid endarterectomy can decrease the five-year risk of ischemic stroke by approximately half. For those with extra-cranial stenosis between 50 and 69%, carotid endarterectomy decreases the 5-year risk of ischemic stroke by about 16%. For those with extra-cranial stenosis less than 50%, carotid endarterectomy does not reduce stroke risk and may, in some cases, increase it. The effectiveness of carotid endarterectomy or carotid artery stenting in reducing stroke risk in people with intracranial carotid artery stenosis is unknown. In carotid endarterectomy, a surgeon makes an incision in the neck, opens up the carotid artery, and removes the plaque occluding the blood vessel. The artery may then be repaired by adding a graft from another vessel in the body or a woven patch. In patients who undergo carotid endarterectomy after a TIA or minor stroke, the 30-day risk of death or stroke is 7%. Carotid artery stenting is a less invasive alternative to carotid endarterectomy for people with extra-cranial carotid artery stenosis. In this procedure, the surgeon makes a small cut in the groin and threads a small flexible tube, called a catheter, into the patient's carotid artery. == Function == Although the function of alpha-synuclein is not well understood, studies suggest that it plays a role in restricting the mobility of synaptic vesicles, consequently attenuating synaptic vesicle recycling and neurotransmitter release. An alternate view is that alpha-synuclein binds to VAMP2 (a synaptobrevin) and stabilizes SNARE complexes; though recent studies indicate that alpha-synuclein–VAMP2 binding is critical for alpha-synuclein-mediated attenuation of synaptic vesicle recycling, connecting the two seemingly divergent views. It may also help regulate the release of dopamine, a type of neurotransmitter that is critical for controlling the start and stop of voluntary and involuntary movements. Alpha-synuclein modulates DNA repair processes, including repair of double-strand breaks (DSBs). DNA damage response markers co-localize with alpha-synuclein to form discrete foci in human cells and mouse brain. Depletion of alpha-synuclein in human cells causes increased introduction of DNA DSBs after exposure to bleomycin and reduced ability to repair these DSBs. Some historians explain the reluctance to declare independence as a "mask of Ferdinand VII": that is, that Patriot leaders felt they needed to claim loyalty to the deposed monarch to prepare the masses for the radical change that full independence would eventually entail.
